What is it?

Palmitoyl Tripeptide-3 (INCI: Palmitoyl Tripeptide-3) — palmitoylated tripeptide. MW: ~600-700 Da. Mechanism: the peptide part activates TGF-beta receptors → SMAD2/3 phosphorylation → activation of transcription of COL1A1, COL3A1 (collagen I and III). Palmitoyl-‘anchor’: increases lipophilicity → better penetration through the lipid layer of the SC → higher bioavailability than that of the pure peptide. Water-oil solubility due to the amphiphilic structure. TGF-beta-like effect without stimulating cell proliferation (safer profile).

Mechanism: binding to TGF-beta receptor II → activation of SMAD2/3 → increased transcription of COL1A1 and COL3A1. In vitro: increased pro-collagen I synthesis by 119% at 1 µM.
